Closest Airport to Port St. Lucie, Florida
Port St. Lucie has no airline airport, so you pick a direction
Port St. Lucie is one of the biggest cities in Florida, and you cannot fly to it. The nearest runway, Treasure Coast International in Fort Pierce, only handles private planes.
That leaves two sensible airports, one in each direction on I-95:

Vero Beach is the closest airport to Port St. Lucie with airline flights. Palm Beach is 15 minutes farther and has many times the choice. For most trips those 15 minutes do not matter, so the better fare should win.
Where you stay shifts it a little. From Tradition or anywhere near I-95 in the south of the city, Palm Beach is a quick run. From St. Lucie West or the north side, Vero Beach is noticeably closer.
Which one I would book, by reason for the trip
Visiting family from the Northeast
Look at Vero Beach first. JetBlue flies there daily from New York (JFK) and Boston, and Breeze flies from several smaller Northeast airports such as Hartford. A nonstop into VRB and a 40-minute drive is hard to beat. If the days or prices do not line up, Palm Beach has plenty of Northeast flights as a fallback. Mets spring training
The Mets train at Clover Park in St. Lucie West from mid-February to late March. From the ballpark, Vero Beach is the closer airport. The problem is that everyone else has the same idea, and VRB has few seats. Book early, or use Palm Beach, where there are more flights to absorb the demand.
A golf trip or a short break
Palm Beach. You will probably want a specific flight time, a direct flight home on Sunday evening and a rental car waiting. PBI gives you the most options for all three.
Coming from the Midwest, the South or the West
Palm Beach again, or a connection through Charlotte into Vero Beach on American. Compare the two. The Charlotte route can be surprisingly quick if the layover is short.
Staying for the winter
If you are down for weeks, price matters more than the drive. Check all the airports in the next section too. One longer drive at each end of a two-month stay is nothing.
If you are willing to drive farther
Three more airports are in range. Melbourne (MLB) is about 70 miles north, roughly an hour and ten minutes. Fort Lauderdale (FLL) is about 95 miles south, an hour and a half when traffic is kind. Orlando (MCO) is about 120 miles, close to two hours on the Turnpike.
Fort Lauderdale and Orlando are where the cheapest fares in Florida usually are, and both have international flights. I would only use them when the saving is clear, around $100 a ticket or more. South of Palm Beach, I-95 gets heavy in the afternoon, so do not plan a tight drive back to a Fort Lauderdale flight.
A warning about March
Spring training, spring break and the end of the snowbird season all land in the same few weeks. Fares into every airport here go up, and rental cars sell out. If your trip is in March, book the flight and the car together, and book them early.
What I would do
Search Vero Beach and Palm Beach side by side for your dates. Pick the cheaper one unless the difference is small, in which case pick the better flight time. Only widen the search to Fort Lauderdale or Orlando if both look expensive.
You will need a car either way. Port St. Lucie is spread out and has little public transport, and no train or bus links it to these airports in a useful way.
